Preclinical data on RVU602, a novel ADC payload, and the ONCO Prime platform to be presented at the 2026 EORTC-NCI-AACR Symposium The Management Board of Ryvu Therapeutics S.A. with its registered office in Kraków _the "Company", "Ryvu"_ announces that the Company will present two posters with preclinical data on RVU602, its proprietary novel ADC payload, and on the ONCO Prime platform at the 2026 EORTC-NCI-AACR Symposium on Molecular Targets and Cancer Therapeutics _ENA_, November 18-20, 2026, Barcelona, Spain. Details on the poster presentations are as follows: Abstract Title: RVU602: A Non-TOP1/Non-Tubulin Cytotoxic and Immunomodulatory Payload for Next-Generation ADCs Abstract Number: 72 Session date and time: Wednesday, November 18 _09:00-17:00 CET_ RVU602 is a proprietary, potentially first-in-class ADC payload. Its mechanism of action is distinct from both topoisomerase I _TOPO1_ inhibitors and microtubule inhibitors, the two payload classes on which nearly all approved and late-stage ADCs rely. RVU602 was discovered on Ryvu's payload discovery platform, which integrates patient-derived models and high-throughput phenotypic screening. RVU602 disrupts an essential intracellular survival process. It shows broad activity across cancer cell states, including those less responsive to classical payloads. In a subset of cell lines, it showed higher potency than DXd _mean EC50 of 34 nM vs. 45 nM_. RVU602 was conjugated to four antibodies: sacituzumab _anti-TROP2_, cetuximab _anti-EGFR_, telisotuzumab _anti-c-MET_ and trastuzumab _anti-HER2_. It formed active ADCs with all of them, reaching sub- to low-nanomolar potency _EC50 of 0.22-14 nM in TROP2-responsive models_, a 3- to 23-fold shift versus the free payload. In TROP2 models, the sacituzumab-RVU602 ADC matched or exceeded datopotamab deruxtecan _Dato-DXd_. The cetuximab-RVU602 ADC was on par with a cetuximab-MMAE comparator. The sacituzumab-RVU602 ADC was efficacious and well tolerated in the TROP2-positive HCC70 xenograft model. Beyond direct cytotoxicity, RVU602 induces the release of extracellular ATP, a hallmark of immunogenic cell death, exceeding the effect of a topoisomerase inhibitor. Together, these data confirm the activity of RVU602 across multiple antibodies and in settings resistant to TOPO1 inhibitors. Validation of tolerability and therapeutic window is ongoing. Abstract Title: Mapping Hidden Vulnerabilities in Genetically Stratified Colorectal Cancer with ONCO Prime Abstract Number: 195 Session date and time: Thursday, November 19 _09:00-17:00 CET_ Ryvu has developed a proprietary platform, ONCO Prime, to identify actionable vulnerabilities in genetically defined cancer contexts. The platform combines clinically relevant model systems, including healthy human intestinal stem cells _hISCs_, engineered isogenic colorectal cancer _CRC_ models carrying key oncogenic drivers such as APC and KRAS alterations, and patient-derived cells _PDCs_. These models are amenable to genetic perturbation and high-throughput pharmacological testing. Molecular profiling, transcriptomic analyses and computational approaches further support patient stratification and link functional phenotypes with pathway states. Using genome-wide CRISPR/Cas9 loss-of-function screening followed by orthogonal validation in patient-derived CRC models, the team identified multiple synthetic lethal _SL_ interactions associated with WNT and RAS/MAPK pathway dysregulation. The platform enabled prioritization of novel targets with first-in-class therapeutic potential. It also revealed opportunities for drug repurposing of clinically advanced or approved agents. In genetically stratified CRC models, selected pharmacological interventions demonstrated robust in vitro activity, including context-dependent responses and combination potential. Together, these findings demonstrate that ONCO Prime can systematically connect cancer genotype, pathway activity and functional vulnerability to nominate therapeutically relevant targets. This supports the development of both first-in-class programs and repositioning strategies for defined CRC patient populations. The posters will be available on the Company's website after the presentations: https://ryvu.com/publications Disclaimer: This English language translation has been prepared solely for the convenience of English-speaking readers.
